This study presents an efficient strategy based on liquid–liquid extraction, high‐speed counter‐current chromatography, and preparative HPLC for the rapid enrichment, separation, and purification of four anthraquinones from Rheum tanguticum. A new solvent system composed of petroleum ether/ethyl acetate/water (4:2:1, v/v/v) was developed for the liquid–liquid extraction of the crude extract from R. tanguticum. As a result, emodin, aloe‐emodin, physcion, and chrysophanol were greatly enriched in the organic layer. In addition, an efficient method was successfully established to separate and purify the above anthraquinones by high‐speed counter‐current chromatography and preparative HPLC. This study supplies a new alternative method for the rapid enrichment, separation, and purification of emodin, aloe‐emodin, physcione, and chrysophanol.  相似文献

The separation of polar compounds by high‐speed countercurrent chromatography is still regarded as a challenge. In this study, an efficient strategy for the separation of three polar compounds from Rheum tanguticum has been successfully conducted by using high‐speed countercurrent chromatography. X‐5 macroporous resin chromatography was used for the fast enrichment of the target compounds. Then, the target fraction was directly introduced into high‐speed countercurrent chromatography for separation using ethyl acetate/glacial acetic acid/water (100:1:100, v/v/v) as the solvent system. Consequently, three polar compounds including gallic acid, catechin, and gallic acid 4‐O‐β‐d ‐(6′‐O‐galloyl) glucoside were obtained with purities higher than 98%. The results showed glacial acetic acid could be such an appropriate regulator for the ethyl acetate/water system. This study provides a reference for the separation of polar compounds from natural products by high‐speed countercurrent chromatography.  相似文献

Anthraquinone glycosides, such as chrysophanol 1‐O‐β‐d‐ glucoside, chrysophanol 8‐O‐β‐d‐ glucoside, and physion 8‐O‐β‐d‐ glucoside, are the accepted important active components of Rheum tanguticum Maxim. ex Balf. due to their pharmacological properties: antifungal, antimicrobial, cytotoxic, and antioxidant activities. However, an effective method for the separation of the above‐mentioned anthraquinone glycosides from this herb is not currently available. Especially, greater difficulty existed in the separation of the two isomers chrysophanol 1‐O‐β‐d‐ glucoside and chrysophanol 8‐O‐β‐d‐ glucoside. This study demonstrated an efficient strategy based on preparative high‐performance liquid chromatography and high‐speed countercurrent chromatography for the separation of the above‐mentioned anthraquinone glycosides from Rheum tanguticum Maxim.ex Balf.  相似文献

青海地道中药材“西宁大黄”野生资源濒临枯竭,人工种植规模不断扩大。定期采集青海西宁及邻近地区两年生种植大黄,跟踪分析了其微量无素铜、锌、铁、锰、铬,为地道西宁大黄资源的可持续开发和利用提供科学依据。研究表明,种植大黄中微量元素铁、锌、铜、锰的有效积累以及土壤地球化学环境中的微量元素锌、锰、铜对种植大黄的生长具有重要的作用和意义。  相似文献

利用超临界流体二氧化碳萃取技术提取中药虎杖、大黄、炒白术中黄酮类化合物。在pH8.2的HCl—tris缓冲溶液中利用邻苯三酚自氧化法测定提取物对超氧阴离子自由基的消除作用。用茜素紫作显色剂的分光光度法在pH9.0的HCl—tris缓冲体系中测定中药提取物对Co^2 H2O2体系所产生的羟基自由基的消除作用,并对实验条件进行了探讨。实验结果表明,3种中药提取物对自由基都有消除作用,其中虎杖的效果最好。  相似文献

An organosilane containing binaphthyl functional group was synthesized by clicking 2,2'-bis(prop-2-ynyloxy)-1,1'-binaphthyl with 3-azidopropyltriethoxysilane (AzPTES). The "click" binaphthyl stationary phase was then synthesized by covalently bonding the organosilane onto silica beads. In this synthetic method, the residue of copper iodide (CuI) catalyst can be controlled at a very low level. Polycyclic aromatic hydrocarbons (PAHs) were well retained on the "click" binaphthyl column, and showed different retention factors (k) and separation ratio (α) values from those on octadecylsilyl (ODS) column due to the π-π interaction between the analytes and the stationary phase. The anthraquinone compounds in Rheum palmatum L. were selectively separated and enriched into a fraction by the "click" binaphthyl column. By using this way, the complexity of the sample was largely reduced. Twelve of anthraquinones were recognized by UPLC-ESI-MS/MS. This stationary phase will be very useful for separating and enriching similar compounds with planar aromatic structures from complex traditional Chinese medicine (TCM) samples.  相似文献

中药大黄多糖中单糖组成的毛细管区带电泳分析   总被引:5,自引:2,他引:5  
以1-苯基-3-甲基-5-吡唑啉酮(PMP)为单糖的衍生化试剂,建立毛细管区带电泳(CZE)同时分离分析8种常见还原单糖PMP衍生物的方法。将该方法用于中药大黄多糖(RTP)的单糖组成及其摩尔比率的测定。结果表明,在pH 10.8和150 mmol/L硼砂缓冲溶液、10kV分离电压、25℃柱温的优化条件下,8种单糖衍生物实现了良好的分离,并证实RTP由阿拉伯糖、葡萄糖、半乳糖、葡萄糖醛酸和半乳糖醛酸5种单糖组成,其摩尔比为8.01∶5.01∶30.30∶1.00∶1.56;样品测定回收率为96.4%~105.3%。该方法灵敏、快速、准确,可用于中药RTP的组成分析。  相似文献

The dried roots of Rheum officinale Baill., named Dahuang in Chinese, has many pharmacological effects and has been widely used for the treatment of many diseases. To develop a harmless and eco-friendly method for the separation of components in Dahuang is of great interest for quality control and pharmacological study of Dahuang. A method for separation and purification of components in Dahuang using supercritical fluid chromatography (SFC) is established in this work. Samples were prepared by extraction of Dahuang powders with 20% H2SO4 and benzene under reflux. The extracts were obtained by evaporating benzene extracts and separating by SFC on YMC-Diol column using supercritical CO2 with CH3OH 4% (v/v) as the modifier. The flow rate of the mobile phase was 15?mL/min, the column pressure was 13?MPa, and the column temperature was 318?K. Cinnamic acid and five kinds of hydroxyanthraquinones including rhein, emodin, aloe emodin, chrysophanol, and physcion were obtained by SFC. The purities of the obtained compounds were all above 97% as determined by high performance liquid chromatography and their chemical structures were identified by nuclear magnetic resonance and mass spectrum. The thermodynamics of chromatographic process was also studied and it revealed that the SFC separation process of these compounds on YMC-Diol column was controlled by enthalpy.  相似文献
